چکیده

The early studies on conjugase from chicken pancreas (1, 2) did not lead to the identification of this enzyme. Subsequent progress in the elucidation of the chemical structure of pteroylglutamic acid (3) and its derivatives (4, 5) left little doubt that conjugase is one of the peptidases. Pfiffner et al. (4) classified the conjugase enzymes as carboxypeptidases after they showed that the methyl ester of pteroylheptaglutamate was not attacked by conjugase, while the free acid was attacked. In the present paper the results of some experiments designed to determine the specificity of conjugase toward several suspected substrates are reported.

Observations on folate absorption with particular reference to folate polyglutamate and possible inhibitors to its absorption.

A test system for detection of inhibitors to the enzyme which removes the glutamic acid peptide chain from folate polyglutamates (folate conjugase) is described. This utilizes the reaction between plasma conjugase and the folate polyglutamate in red blood cell haemolysate. Diphenylhydantoin and yeast extracts did not inhibit plasma conjugase. The better absorption of monoglutamate forms of fola...

Inhibition of folate enzymes by sulfasalazine.

Sulfasalazine (salicylazosulfapyridine), an agent widely used for the treatment of ileitis and colitis, is also a competitive inhibitor of intestinal folate transport (1, 2). The mechanism of action of sulfasalazine remains uncertain. To further explore the mechanism of sulfasalazine action, the interaction of the drug with the folate recognition site was tested with three enzymes: dihydrofolat...

P102: The Association of the Anti-GAD Antibodies to the Neurological Conditions

Glutamic acid decarboxylase (GAD) is an enzyme which converts the glutamic acid to the neurotransmitter gamma-amino butyric acid (GABA). GABA is an inhibitory neurotransmitter that inhibits or weakens the neuronal stimulations. Presynaptic GABAergic neurons in the central neurons system (CNS) and the cells in the islets of Langerhans in the pancreas generate GAD. There are two isoforms of GAD n...

[Tetrazolium bioautography of pteroylglutamic acid conjugates].

A microbioautographic method for the identification of pteroylglutamic acid derivatives and their glutamyl conjugates is described. Chromatograms or pherograms of mixtures of the compounds are treated with chicken pancreas conjugase, followed by tetrazolium bioautography. This combined procedure allows qualitative and semi-quantitative analyses of folate compounds in biological material in the ...
